Answer
Yes. The 2 damage already marked combines with the blocker's 2 combat damage, so both creatures die.
Why
The 4/4 starts this dispute with 2 damage already marked on it. Combat adds the blocker's 2 damage, bringing the total marked damage to 4.
Damage stays marked until cleanup. When state-based actions see damage equal to the 4/4's toughness, they destroy it; the 4 damage it dealt also destroys the 2/2.
